Everyone in the main 6 compliments one another quite nicely. This pairing in particular is a huge case of the prankster and the seriousster. Rainbow doesn't take much stuff seriously, while Twilight's a pretty big 'by-the-rules' kind of girl. That would be their conflicting point. And that would affect their relationship minimally, I think. This is also a great standing point for their relationship. They'd balance each other.
By preference;
It's not something I'd ship normally, but I wouldn't be bothered if it appeared as a side pairing in a fic. Neutral ground.
