Jillian Petrie Octavius
- Doctor Otto Octavius's daughter
- Deceased mother, was killed by a murderer drowning her (Jillian had a chance to stop him before the crime, but didn't)
- Best friends to Peter and Harry Osborn, Mary Jane Watson, and Gwen Stacy
- Attends Horizon High, but then is later transferred to Midtown Tech
- Attends Empire State University for college
- Still one year younger than Peter but is in the same school year
- Is 14 when she is first bitten by the radioactive spider
- Is first in a relationship with Harry, but then he becomes the Hobgoblin and sacrifices himself
- Ends up with Flash Thompson after her second year of college

Peter Benjamin Osborn
- Norman Osborn's son, Harry Osborn's older brother by one year
- Deceased mother, was killed in an arson fire (Peter had a chance to stop him before the crime, but didn't)
- Best friends to Jillian Octavius, Mary Jane Watson, and Gwen Stacy
- Attends Midtown High
- Attends Empire State University for college
- Is 15 when he is first bitten by the spider
- Has a crush on Gwen Stacy, but she turns him down after learning of his hero alter ego
- End up with MJ at the start of college

Spiderlings' origin story
Peter invites Jillian over to his house often because they are inseparable best friends. They always work on projects together while Harry is out doing social things. One day, Peter and Jillian agree to get some things from Peter's dad's lab. Even though he isn't allowed down there, Peter gets them access and they sneak inside. While down there, Jillian gets bit by a spider that got out of its test tube. She freaks out and flicks it onto Peter who also gets bitten. They run upstairs and lock the lab to keep Norman from ever knowing. They develop spider powers a few days later and decide to tell Harry about it. On their way home, they run into one of Norman's business rivals robbing a bank. They let him pass and don't do anything to stop him. The two go to dinner instead of heading home right away, and ultimately decide to tell Harry in the morning. When Jillian gets back to her house, she discovers that her mother was killed by drowning. Peter finds out that his own mom was killed in an Osborn arson fire. The two meet up and figure out that their mothers were at the same building the night of their deaths. Someone started a fire, which killed Mrs. Osborn. Mrs. Octavius escaped, but was caught by the arson and murdered in the Hudson since it was right by the building that burned. After listening to the police radios, Jillian and Peter find the criminal to see it was the Osborn rival they ran into but didn't stop. They then swear to use their powers to help people. Even if their fathers turn into the Green Goblin and Doctor Octopus later on, they stick true to their morals and become the one and only Superlative Spiderlings!

May and Benjamin Parker
- Not blood related to Jillian or Peter
- They live normal lives and give extra money to the nearby high schools
- Sometimes attend Midtown Tech sports games
- Acquaintances of Jillian and Peter (they ran into them at some point when the two were late for school and gave them a ride)

Sarah Green
- Young and innovative technician of Stark Industries
- Was offered a place at OSCORP but denied it
- Helped create the foresight Iron Man suit (but only sees the field once before it's destroyed)
- 28 years old and graduate of MIT college and a high school in Kansas
- Usually fights alongside Iron Man or chills with Jessica Jones
- Helps the Spiderlings out with their Iron Suits
- Unaware of their identities but has a hunch they have connections with the Osborns and Octavius
- Engaged to Quinten Beck

Gwen Stacy
- Student at Horizon High
- Same age as Peter, besties to Jillian and Mary Jane
- Does not get spider powers in this universe, but loves to help the Spiderlings as Gwen (with the power of her dad being a chief of police)
- Figures out their identities early on

Mary Jane Watson
- Student at Midtown
- Besties to Jillian, Peter's main love interest
- Works as a waitress in college, wants to be an actress for Beck Theaters until it turns out the owner is Mysterio
- Gwen tells her Peter is Spider-Man after an argument

Flash Thompson
- Midtown High's QB in football
- Athletic, appears to be dumb but is really intelligent
- The Spiderlings' biggest fan
- Halfway through discovers their identities after seeing Jill's web formula in her lab locker in college
- Attends Empire State with Jill and Peter after graduating high school

Harry Osborn
- Peter's younger brother
- Jill's main love interest until just after they reach college
- Becomes Hobgoblin and later sacrifices himself to save Spider-Woman from being cloned by the Jackal
- Attends Midtown and later Empire State until he dies by the Jackal's hand
